Two rat and one human model systems will be used: (1) the isolated rat lens epithelial cell culture in monolayer; and (2) the intrastromal lens culture in organ culture. A partial purification of lens epithelium-derived growth factor (LEDGF), a nuclear protein with affinity for the Krex transcription factor, has been achieved. It has been shown that LEDGF induces proliferation of lens epithelial cells, binds to the a-sag protein on the cell membrane, and is an important target for the action of interleukin-1 on these cells. The specific aims of the project are: (1) to determine the effect of alcohol and acetaldehyde on lens epithelium-derived growth factor; (2) to determine if the toxicity of alcohol and acetaldehyde are mediated through the redox component; (3) to determine if the toxicity is mediated through the a-sag component; (4) to define the mechanism of the effect of alcohol and acetaldehyde on the cytoskeleton of lens epithelial cells; (5) to evaluate the preventive effect of LEDGF on lens epithelial cell toxicity.
