The medial nasal fold of the nasal epithelium develops by an endoderm-mesenchymal interaction during embryonic development. Fgf-8, a member of the fibroblast growth factor (FGF) family, is expressed in the periderm of the surface ectoderm and developing mesenchyme. In order to clarify the precise role of Fgf-8 in the development of the medial nasal fold, we established Fgf-8-deficient mice. Histological and immunohistological analyses revealed that the medial nasal epithelial fold failed to develop in mutant mice, although the ventral surface epithelium was present and followed the same developmental path as that in the wild-type embryo. This suggests that Fgf-8 is essential for the development of the medial nasal fold. In wild-type embryos, a Fgf-8-expressing mesenchymal cell sheet was observed to rise toward the nasal ectoderm. This was found in all Fgf-8-deficient embryos, and suggested that the mesenchyme was inserted into the nasal epithelium. RT-PCR analysis revealed that the mesenchyme of Fgf-8-deficient mice expressed the same level of Tbx-5 and Tcf-4 as the mesenchyme of the wild-type embryo. Histological and immunohistological analyses, however, failed to detect any abnormal Tbx-5 and Tcf-4 expression by the nasal mesenchyme.
