Complement factor C3 is the most abundant of the complement components in serum, where it is converted to its soluble C3a and C3b form by two regulated proteases; thrombin-like and transglutaminase-like enzymes. The production of C3a and C3b leads to the generation of various peptide fragments (C3a, C3b, C3dg). C3a peptides are produced by proteolytic cleavage of the amino terminus and carboxy terminus of C3. The function and biological activity of C3a, C3b and C3dg peptides in vivo are not known. We show that injection of the C3aR antagonist HS-C3-6 (1 microg) before injection of C3 knockout mice with apoptotic thymocytes does not affect the subsequent splenic clearance of apoptotic cells, unlike neutralization of C3a by HS-C3-6 (1 mg) or C3-derived peptide 10 (500 microg) which reduces clearance. Injection of C3 knockout mice with C3-derived peptide 10 led to increased splenic clearance of apoptotic cells when compared to injection with thymocytes alone. Injection of C3-derived peptide 10 also reduced clearance of apoptotic leukocytes. Administration of C3-derived peptide 3 (100 microg) led to increased splenic clearance of apoptotic cells in C57BL6/J mice. Finally, intravenous injection of C3-derived peptide 10

A 2-year-old Japanese girl had multiple bone fractures, dentinogenesis imperfecta, short stature, thinness, and bone deformities. Radiographs showed fractures of both femoral necks, metacarpals, ribs, tibiae, and phalanges, bone thinning, and looser joints. The patient had mild facial deformities and muscle weakness. Body shape was concave but not short. The fracture-line was poorly sclerotic. Physical findings of the patient were remarkable for marked bone deformities, multiple fractures, short stature, and bone fragility. The coefficient of variation of z-scores on bone mineral density of the lumbar spine was 36% and of the whole body was 44%. The coefficient of variation on the grading of Sillence was 50%. This report defines a new form of type IIA osteogenesis imperfecta.
